Inhibitory Effect of Deep Sea Water Containing Chitosan Oligosaccharides on Adipocyte Differentiation

Abstract
To determine effect of deep sea water (DSW) containing chitosan oligosaccharides (COS) on adipocyte differentiation, we performed Oil Red O staining to visualize and quantify lipid accumulation in differentiated 3T3-L1 adipocytes with the treatment of DSW alone or DSW containing COS. Both adipocyte differentiation and lipid accumulation were inhibited with the DSW alone or DSW containing COS in 3T3-L1 adipocytes in a dose-dependent manner. The amount of triglyceride (TG), an ester derived from glycerol and free fatty acids, in 3T3-L1 adipocytes was also decreased. Furthermore, expression of adipogenic genes: peroxisome proliferator-activated receptor gamma (PPAR-¥ã) and CCAAT/enhancer-binding protein alpha (C/EBP ¥á), major adipogenic transcription factors, and glucose transporter 4 (GLUT4), a mature adipocyte marker, was decreased. These data suggest that DSW containing COS is more effective than DSW alone in inhibitory effect on adipocyte differentiation.
